一、 简历: 1981-1985 安徽师范大学生物系 学士 1985-1988 广西农学院分子遗传学研究室 硕士 1985-1986 华东师范大学生物系(联合培养) 硕士 1989-1992 英国John Innes Centre 博士 1992-1998 英国剑桥大学植物科学系 博士后 1998-至今 中国科学院国家基因研究中心 主任,研究员 2002-至今 中国科学院上海生科院植物生理生态所 副所长 2008-至今 中国科学院北京基因组研究所 副所长 二、 所获学术荣誉、奖项 2003.3 中国科学院创新文化建设先进个人 2003.3 中科院2001-2002年度重大创新贡献团队 2003.9 留学回国人员成就奖(中组部、人事部和科技部等) 2003.12 上海市科学技术进步一等奖奖 2004.4 2001-2003年度上海市劳动模范 2004.10 中国科学院优秀研究生指导教师 2007.12 国家自然科学二等奖 三、 主要学术成果 长期从事基因组学的基础研究,并致力于利用不断发展的基因组学的理论和方法探索水稻基因组结构、功能和遗传改良潜力。完成了对水稻第四号染色体的精确测序和综合分析,发展了籼、粳稻全基因组及基因水平上的比较与功能研究系统,建立了以第二代测序仪为基础的高通量基因型分析平台。有关研究结果已发表在Nature, Genome Research, Plant Cell等国际学术刊物上。 1. 水稻4号染色体精细物理图的构建及基因组精确测序 通过整合分子标记、克隆末端序列、DNA指纹等数据,运用分子、染色体荧光原位杂交和比较基因组分析的方法,构建了粳稻4号染色体高精确度和高覆盖率(98.7%)的物理图。鉴定了4号染色体的物理长度以及物理图距和遗传图距的对应关系。基于染色体精细物理图,采用克隆连克隆法的基因组测序方法,完成水稻4号染色体的精确测序和分析。研究成果发表在2002年11月21日出版的Nature上。 之后继续承担了水稻全部12条染色体着丝粒序列和结构的分析,以及籼、粳稻全基因组序列比较,直到2005年国际水稻基因组测序计划完成。 2. 水稻4号染色体结构及功能的深入研究 在精细物理图构建和精确测序的基础上,突破了拼装高度重复序列的技术难题,成功鉴定了覆盖着丝粒区域的大片段克隆,完成了序列测定和组装。发现水稻4号染色体着丝粒核心区是由近360个高度重复的特异序列组成,并将这些重复序列组装成18个串联式重复区段,还在该区域鉴定一些编码基因。 在精确测序的基础上,研制了水稻4号染色体的覆瓦式排列的特异芯片,并通过芯片杂交分析,开展了籼、粳稻基因比较表达谱分析和转录调控因子的研究。鉴定了水稻4号染色体上一批新的表达基因,比较了籼粳基因功能和调控方面的差异。得出了水稻染色体上相邻排列的基因有很高的协同表达调控特征等重要结论。还印证了之前对4号染色体基因分布规律的推测结果。 3. 比较基因组和功能基因组研究 通过对籼、粳稻基因组的比较分析,系统鉴定了籼粳之间序列多态性分布和频率。随着水稻基因组精确测序的完成,建立水稻转基因实验室,建设了一定规模的温室和试验田。高通量研究水稻基因功能,对一万多条籼稻全长cDNA进行了克隆,测序和分析。同时,通过比较基因组学研究了转座子突变可能对水稻基因功能分化产生的影响。结合基因家族的比较分析和表达组、转基因、过表达等功能分析,发现了一系列在水稻抗热,抗旱和抗盐生理过程中起重要作用的基因。 4. 建立以第二代测序仪为基础的高通量功能基因组研究平台 最近,新一代DNA测序方法的诞生标志着生命科学技术的又一次飞跃,为大规模发现和利用水稻遗传资源开辟了有效的新途径。这种新方法集低时耗、高性价比、高密度和高精度等众多优点于一身。采用这一方法完成了一组籼、粳稻重组自交系的基因型鉴定,与目前广泛应用的分子标记相比,新方法在速度和精度上有了大大提高。其精度是之前传统的方法手段无法实现的。 同时尝试的高通量功能基因组研究的新技术还包括表达组和染色质免疫沉淀测序。迅速、大量地鉴定覆盖全基因组的单核苷酸多态性,全面深入掌握我国水稻资源的遗传多样性。
